openHCA is our approach to open high content analysis based on open automated microscopy hardware designs and utilising open source software for data acquisision and analysis. We are working to implement advanced microscopy techniques (e.g. FLIM, FRET, OPM, QPM, STORM) for unsupervised imaging of sample arrays (e.g., in multiwell plates) to enable the routine interrogation of 100s of fields of view under well-controlled environmental conditions. The schematic below indicates the key components of an HCA system. The components in green font are already prototyped in our laboratory and those in orange font are components that we purchase. We initially developed HCA systems using commercial motorised fluorescence microscope frames that incorporated an optical autofocus module. Subsequently we have developed an open optical autofocus module that we can implement on a commercial microscope frame or on an openFrame microscope system.
